TEM样品无铀染色-醋酸双氧铀的替代品
UranyLess TEM staining alternative for Uranyl Acetate
荷兰Micro to Nano公司（简称M2N）提供的Uranyless无铀染液对TEM染色应用而言，是一种全新的提高对比度的染色试剂，可以取得和醋酸双氧铀一样的实验结果，且不会产生与醋酸双氧铀相关的限制、毒性和污染物清理等问题。
无铀染液
Uranyless无铀染液是多种镧系元素的专用混合物，对生物材料具有很大的亲和力，是一种可直接使用的水溶液成品。它已经在动物和植物的许多生物组织上进行了染色测试。无铀染液是超细切片和负染色的理想选择。该无铀染液是一种高效的、无放射性的镧系稀土染色混合物。无辐射担忧，染色速度快，仅接触一分钟后，切片或沉积物就可被染色，从而为后续TEM成像提供对比度。如果需要，根据雷诺兹经典染色方案（1963年），建议使用柠檬酸铅来增强对比度。
无铀染液采用无空气瓶包装，容量为30ml。无空气瓶在没有空气的情况下灌装染液，并通过按压泵装置输送溶液的液滴。这消除了二氧化碳污染。对于30毫升包装，你可以染大约1500个TEM载网。无空气瓶中的无铀染液在打开后的保质期为12个月，通过输送液滴避免浪费。无铀染液的pH值为6.8至7。可选包装容量为200ml，用于徕卡EM染色自动染色机。
柠檬酸铅
尽管无铀染液已具有很强的对比度效果，但根据雷诺兹经典染色方案，推荐使用柠檬酸铅来进一步增强对比度。我们提供一种3%浓度的柠檬酸铅即用溶液，采用无空气瓶包装，在没有空气的情况下灌装入瓶，通过按压泵送装置输出柠檬酸铅液滴，从而消除二氧化碳污染。柠檬酸铅溶液作为高电子密度着色剂广泛用于电镜实验室，适于超薄切片着色，经常用于铀先着色之后再提高对比度，且用于多种细胞和组织成分着色。
染色流程
1.滴一滴无铀染液在聚乙烯保鲜膜或疏水载玻片上
2.将TEM载网放置在无铀液滴上浸透1-2分钟
3.用滤纸吸干TEM载网
4.然后用蒸馏水清洗TEM载网
5.将装载样品的TEM载网晾干
